I would add it before I add the water.

I like to add it as I go but I’ve also added it before and after with no ill effects.

I always do it before.

I would never add untreated water to a tank, I always pre mix my water in a drum first, even for my 100 gallon, and I change 75% of the water once a week in all my tanks.

I would do it before so that the flow of the new water can mix with the Prime.

I put it before I started refilling. If you really wanted to try to optimize it, might be worth dumping in before and after but I think before is better.

Directions say to put in amount for total gallons (not just water you are replacing) so the reaction time is faster. I suppose one could dump say 75% of that dose before and the rest when you're almost done but that's probably just unnecessarily complicating things.

Again, directions say to add amount for volume of c tank and then fill up.

I've had 20+ tanks for several years and do water changes twice a week. I fill the tanks with a hose directly from the tap. I add Prime just before I fill with tap water. I've never had any problem with water changes.

So, that was all helpful eh? You can add the Prime before, during or after you add the water. The instructions say they recommend pre-treating the water you add but that it can be added directly to the tank. In that case, you add enough to treat the entire tank volume, not just the amount being replaced.
So, it's your preference how you do it, looks like!
