I currently have a 45g, and will be upgrading to a 75g soon, and plan to reuse the sand. Before I transfer the sand, should/can I wash it in a bucket to get rid of all the algae, fish waste, and detritus?

I would, no point transferring unwanted gunk.

Yeah, new water, new everything. Current water has a slight tint of brown from the driftwood aswell.

rinse it lightly to get the large clumps of waste out, but not all and not too well. when its too clean youll get a white cloudy bloom. save some brown water with solids, itll make the water crystal clear.
