I don't know to much about mudskippers. All I'm sure of is yes, they do need land. Hopefully someone else can help you out with that, someone here must have them.
You'll need a half and half tank with a nice sloped ramp for them to use to get on dry ground...

How deep mud do you intend? How would you keep it so mud doesnt create aearobic activities to a point it mught produce rotten odors? How do you intend to separate from water?
Can you collect mud yourself? How about Mangrove Tree Stems?
